C\u2019est l\u2019\u00e9chantillon qui pose probl\u00e8me. Un \u00e9chantillon mal pr\u00e9lev\u00e9 dans un lot s\u00e9par\u00e9, un pr\u00e9l\u00e8vement de surface dans un r\u00e9cipient o\u00f9 des particules grossi\u00e8res ont flott\u00e9 \u00e0 la surface, ou un \u00e9chantillon contamin\u00e9 par l\u2019humidit\u00e9 lors de sa manipulation peuvent donner l\u2019impression qu\u2019un processus stable et bien ma\u00eetris\u00e9 est hors sp\u00e9cifications.<\/p>\n\n\n\n<p>La cons\u00e9quence pratique est bien r\u00e9elle\u00a0: les ing\u00e9nieurs ajustent les param\u00e8tres de processus sur la base de donn\u00e9es erron\u00e9es. Une mesure erron\u00e9e indiquant une granulom\u00e9trie trop grossi\u00e8re entra\u00eene une augmentation inutile de la vitesse du classificateur ou une prolongation du temps de broyage. \u00c0 l\u2019inverse, une mesure erron\u00e9e indiquant une granulom\u00e9trie trop fine provoque l\u2019effet inverse. Dans les deux cas, ces modifications de processus superflues consomment de l\u2019\u00e9nergie, font perdre du temps et introduisent une variabilit\u00e9 r\u00e9elle dans un processus qui aurait pu \u00eatre parfaitement stable.<\/p>\n\n\n\n<p>Cet article explique pourquoi il est intrins\u00e8quement difficile d&#039;obtenir un \u00e9chantillon repr\u00e9sentatif de poudres s\u00e8ches. Le m\u00e9canisme diff\u00e8re selon le contexte\u00a0: dans un r\u00e9cipient statique, les fines s&#039;infiltrent vers le bas entre les particules grossi\u00e8res sous l&#039;effet de la gravit\u00e9, se concentrant au fond tandis que les particules grossi\u00e8res remontent \u00e0 la surface. Dans un flux transport\u00e9 par air comprim\u00e9, les fines sont entra\u00een\u00e9es pr\u00e9f\u00e9rentiellement par le courant d&#039;air vers les zones mortes et les r\u00e9gions \u00e0 faible vitesse. Parall\u00e8lement, les particules grossi\u00e8res convergent plus directement vers le point de collecte principal. Dans un r\u00e9cipient vibrant (comme lors du transport), les fines migrent vers le bas et les particules grossi\u00e8res remontent \u2014 c&#039;est l&#039;effet de la noix du Br\u00e9sil.<\/p>\n\n\n\n<p>En pratique, cela signifie qu&#039;aucun point de pr\u00e9l\u00e8vement unique dans un lot ne repr\u00e9sente la distribution granulom\u00e9trique compl\u00e8te de celui-ci. Un \u00e9chantillon pr\u00e9lev\u00e9 en surface sera enrichi en particules grossi\u00e8res\u00a0; un \u00e9chantillon pr\u00e9lev\u00e9 au fond sera enrichi en particules fines. Ces deux pr\u00e9l\u00e8vements ne sont pas erron\u00e9s \u2013 il s&#039;agit de mesures r\u00e9elles \u2013 mais aucun ne repr\u00e9sente la distribution granulom\u00e9trique globale du lot telle qu&#039;elle sera per\u00e7ue lors du processus en aval.<\/p>\n\n\n\n<h3 class=\"wp-block-heading\" id=\"h-surface-area-static-charge-and-agglomeration\"><mark style=\"color:#0693e3\" class=\"has-inline-color\">Surface sp\u00e9cifique, charge statique et agglom\u00e9ration<\/mark><\/h3>\n\n\n\n<p>Les poudres fines et s\u00e8ches \u2014 en particulier celles dont le D50 est inf\u00e9rieur \u00e0 10 microns <a href=\"\/fr\/products\/fluidized-bed-jet-mill-supplier\/\" data-internallinksmanager029f6b8e52c=\"9\" title=\"Moulin \u00e0 jet\" target=\"_blank\" rel=\"noopener\">fraisage au jet<\/a> Le fraisage par impact fin, ou le broyage fin, conf\u00e8re aux particules une surface sp\u00e9cifique \u00e9lev\u00e9e et favorise l&#039;accumulation de charges \u00e9lectrostatiques. Ces deux propri\u00e9t\u00e9s entra\u00eenent l&#039;adh\u00e9rence des particules aux parois du r\u00e9cipient, \u00e0 la surface de l&#039;outil de pr\u00e9l\u00e8vement et entre elles. Lors du pr\u00e9l\u00e8vement d&#039;un \u00e9chantillon, les particules les plus fines adh\u00e8rent \u00e0 la surface de la pelle et ne sont pas transf\u00e9r\u00e9es proportionnellement dans le r\u00e9cipient. L&#039;\u00e9chantillon est donc plus grossier que le mat\u00e9riau en vrac.<\/p>\n\n\n\n<p>L&#039;agglom\u00e9ration introduit une autre source d&#039;erreur. Les forces de van der Waals et l&#039;attraction \u00e9lectrostatique entre les fines particules forment des agglom\u00e9rats mous \u2014 des amas de particules qui se comportent comme une seule particule plus grosse lors de la mesure par diffraction laser si les conditions de dispersion ne sont pas ad\u00e9quates pour les d\u00e9sagr\u00e9ger. Utiliser un \u00e9chantillonnage multipoint, jamais un \u00e9chantillonnage de surface.<\/mark><\/h3>\n\n\n\n<p>Le pr\u00e9l\u00e8vement d&#039;un seul \u00e9chantillon en surface d&#039;un sac, d&#039;un bac ou d&#039;un contenant de produit repr\u00e9sente l&#039;erreur d&#039;\u00e9chantillonnage la plus fr\u00e9quente lors du traitement des poudres s\u00e8ches. La surface est syst\u00e9matiquement enrichie en particules grossi\u00e8res par s\u00e9gr\u00e9gation gravitationnelle. Le seul \u00e9chantillon repr\u00e9sentatif d&#039;un contenant statique est celui qui inclut des mat\u00e9riaux provenant de diff\u00e9rentes profondeurs (haut, milieu et bas), combin\u00e9s proportionnellement au volume du contenant \u00e0 chaque niveau.<\/p>\n\n\n\n<p>Pour un sac ou un petit r\u00e9cipient, ins\u00e9rez un \u00e9chantillonneur (un tube creux avec une ouverture lat\u00e9rale) verticalement jusqu&#039;au fond, puis pr\u00e9levez un \u00e9chantillon. Combinez les \u00e9chantillons pr\u00e9lev\u00e9s \u00e0 au moins trois endroits diff\u00e9rents dans le r\u00e9cipient. Pour un tas sur un convoyeur ou au sol, pr\u00e9levez des \u00e9chantillons en surface, dans les couches interm\u00e9diaires et \u00e0 la base \u00e0 l&#039;aide d&#039;un \u00e9chantillonneur tubulaire plut\u00f4t que d&#039;une pelle de surface.<\/p>\n\n\n\n<h3 class=\"wp-block-heading\" id=\"h-2-sample-across-the-full-width-of-a-moving-stream\"><mark style=\"color:#0693e3\" class=\"has-inline-color\">2. \u00c9chantillonnage sur toute la largeur d&#039;un cours d&#039;eau en mouvement<\/mark><\/h3>\n\n\n\n<p>L&#039;\u00e9chantillon le plus repr\u00e9sentatif d&#039;une ligne de production est un \u00e9chantillon transversal pr\u00e9lev\u00e9 sur toute la largeur du flux de poudre \u00e0 sa sortie d&#039;un convoyeur, d&#039;un classificateur ou d&#039;un broyeur. Cette m\u00e9thode de r\u00e9f\u00e9rence capture la distribution granulom\u00e9trique compl\u00e8te \u00e0 la sortie du processus, avant toute s\u00e9paration ult\u00e9rieure. Concr\u00e8tement, cela se traduit par un \u00e9chantillonneur mobile qui parcourt le flux de mani\u00e8re r\u00e9p\u00e9t\u00e9e et pr\u00e9l\u00e8ve un \u00e9chantillon moyen dans le temps.<\/p>\n\n\n\n<p>L&#039;\u00e9chantillonnage en ligne dans une conduite ou un conduit, \u00e0 l&#039;aide d&#039;une sonde \u00e0 entr\u00e9e lat\u00e9rale, est moins fiable car les particules de diff\u00e9rentes tailles ont des vitesses et des trajectoires diff\u00e9rentes dans le flux. Une sonde plac\u00e9e \u00e0 un point fixe dans une conduite capture pr\u00e9f\u00e9rentiellement les particules pr\u00e9sentant certaines caract\u00e9ristiques a\u00e9rodynamiques plut\u00f4t qu&#039;une section transversale repr\u00e9sentative.<\/p>\n\n\n\n<h3 class=\"wp-block-heading\" id=\"h-3-use-a-rotary-sample-divider-for-sub-sampling\"><mark style=\"color:#0693e3\" class=\"has-inline-color\">3. Utiliser un diviseur d&#039;\u00e9chantillons rotatif pour le sous-\u00e9chantillonnage<\/mark><\/h3>\n\n\n\n<p>Une fois l&#039;\u00e9chantillon pr\u00e9lev\u00e9 en plusieurs points, il est g\u00e9n\u00e9ralement n\u00e9cessaire de le r\u00e9duire \u00e0 la quantit\u00e9 requise pour l&#039;analyse instrumentale (de quelques grammes \u00e0 quelques dizaines de grammes pour la plupart des diffractom\u00e8tres laser). Le pr\u00e9l\u00e8vement manuel d&#039;un sous-\u00e9chantillon introduit une \u00e9tape de tri suppl\u00e9mentaire\u00a0: les particules fines pr\u00e9sentes dans l&#039;\u00e9chantillon sont r\u00e9parties de mani\u00e8re in\u00e9gale, et un pr\u00e9l\u00e8vement al\u00e9atoire ne permettra pas de les capturer proportionnellement.<\/p>\n\n\n\n<p>Un diviseur d&#039;\u00e9chantillons rotatif (diviseur \u00e0 riffles) divise de mani\u00e8re constante une poudre en fractions \u00e9gales en r\u00e9partissant le flux descendant sur plusieurs goulottes de collecte. Le sous-\u00e9chantillon issu d&#039;un diviseur rotatif est statistiquement \u00e9quivalent \u00e0 l&#039;\u00e9chantillon initial. Ce n&#039;est pas le cas d&#039;un sous-\u00e9chantillon pr\u00e9lev\u00e9 manuellement \u00e0 l&#039;aide d&#039;une pelle, m\u00eame r\u00e9alis\u00e9 avec le plus grand soin.<\/p>\n\n\n\n<h3 class=\"wp-block-heading\" id=\"h-4-check-for-hard-vs-soft-agglomerates-before-choosing-dispersion-method\"><mark style=\"color:#0693e3\" class=\"has-inline-color\">4. V\u00e9rifier la nature des agglom\u00e9rats (durs ou mous) avant de choisir la m\u00e9thode de dispersion<\/mark><\/h3>\n\n\n\n<p>Les agglom\u00e9rats pr\u00e9sents dans les poudres s\u00e8ches se r\u00e9partissent en deux cat\u00e9gories n\u00e9cessitant un traitement diff\u00e9rent. Les agglom\u00e9rats mous, maintenus ensemble par attraction \u00e9lectrostatique ou par de faibles forces de van der Waals, peuvent \u00eatre d\u00e9sagr\u00e9g\u00e9s par l&#039;unit\u00e9 de dispersion pneumatique d&#039;un diffractom\u00e8tre laser \u00e0 pression standard (1 \u00e0 3 bars). Il convient de v\u00e9rifier la coh\u00e9rence des r\u00e9sultats obtenus avec l&#039;instrument pour diff\u00e9rentes pressions d&#039;injection\u00a0: si la distribution granulom\u00e9trique s&#039;affine sensiblement lorsque la pression de dispersion augmente de 1 \u00e0 3 bars, cela indique la pr\u00e9sence d&#039;agglom\u00e9rats mous et la mesure doit \u00eatre effectu\u00e9e \u00e0 la pression la plus \u00e9lev\u00e9e.<\/p>\n\n\n\n<p>Les agglom\u00e9rats durs \u2014 maintenus par frittage, liaisons chimiques ou forte imbrication m\u00e9canique \u2014 ne peuvent \u00eatre d\u00e9sagr\u00e9g\u00e9s par dispersion pneumatique classique. Ils n\u00e9cessitent un traitement ultrasonique en milieu liquide avant mesure \u00e0 sec, ou une diffraction laser en milieu humide assist\u00e9e par ultrasons. Toute tentative de mesure d&#039;une poudre d&#039;agglom\u00e9rat dur par simple dispersion \u00e0 sec donnera syst\u00e9matiquement une distribution granulom\u00e9trique erron\u00e9ment grossi\u00e8re.<\/p>\n\n\n\n<h3 class=\"wp-block-heading\" id=\"h-5-seal-samples-immediately-after-collection\"><mark style=\"color:#0693e3\" class=\"has-inline-color\">5. Sceller les \u00e9chantillons imm\u00e9diatement apr\u00e8s le pr\u00e9l\u00e8vement<\/mark><\/h3>\n\n\n\n<p>De nombreuses poudres s\u00e8ches \u2014 excipients pharmaceutiques, min\u00e9raux de qualit\u00e9 alimentaire, produits chimiques fins hygroscopiques \u2014 absorbent rapidement l&#039;humidit\u00e9 de l&#039;air ambiant. Quelques minutes d&#039;exposition \u00e0 l&#039;air libre dans un environnement humide suffisent \u00e0 cr\u00e9er une humidit\u00e9 superficielle mesurable, favorisant l&#039;agglom\u00e9ration et augmentant ainsi la valeur D50 mesur\u00e9e. Le r\u00e9cipient contenant l&#039;\u00e9chantillon doit \u00eatre scell\u00e9 imm\u00e9diatement apr\u00e8s le pr\u00e9l\u00e8vement, et non quelques minutes plus tard.<\/p>\n\n\n\n<p>Utilisez des r\u00e9cipients secs et pr\u00e9alablement nettoy\u00e9s. Si l&#039;environnement de production est humide (humidit\u00e9 relative sup\u00e9rieure \u00e0 60 \u00b0C), envisagez de transf\u00e9rer les \u00e9chantillons dans un dessiccateur portable ou d&#039;utiliser des r\u00e9cipients scell\u00e9s sous atmosph\u00e8re d&#039;azote pour cette \u00e9tape. Ces pr\u00e9cautions sont courantes dans l&#039;industrie pharmaceutique et il est conseill\u00e9 de les adopter pour toute poudre fine hygroscopique, quel que soit le secteur d&#039;activit\u00e9.<\/p>\n\n\n\n<h3 class=\"wp-block-heading\" id=\"h-6-mix-before-sub-sampling-but-gently\"><mark style=\"color:#0693e3\" class=\"has-inline-color\">6. M\u00e9langer avant le sous-\u00e9chantillonnage, mais en douceur<\/mark><\/h3>\n\n\n\n<p>M\u00eame un \u00e9chantillon en vrac pr\u00e9lev\u00e9 correctement en plusieurs points pr\u00e9sentera une certaine h\u00e9t\u00e9rog\u00e9n\u00e9it\u00e9 due aux variations locales. Le m\u00e9lange de l&#039;\u00e9chantillon avant le sous-\u00e9chantillonnage r\u00e9duit cette h\u00e9t\u00e9rog\u00e9n\u00e9it\u00e9.
